Spatio-temporal expression of miRNAs in tomato tissues upon Cucumber mosaic virus and Tomato aspermy virus infections.

چکیده
MicroRNAs (miRNAs) play vital roles in regulating plant growth and development. Recent work has shown that miRNA-mediated regulation of cellular mRNA expression is involved in pathogen-host interactions. However, knowledge about the timing and spatial regulation of plant miRNA expression is still limited. Here, we use stem-loop real-time reverse transcription-polymerase chain reaction to quantify the expression changes of seven miRNAs and their target mRNAs in different tomato tissues during the pathogenic processes. Compared with mock-inoculated plants, the expression levels of investigated miRNAs and mRNAs were enhanced by different degrees upon Cucumber mosaic virus (CMV)-Fny and Tomato aspermy virus-Bj infections, but were almost unchanged in CMV-FnyΔ2b (a CMV-Fny 2b-deletion mutant)-infected tomato seedlings. In addition, the obvious up-regulation of several miRNAs and target mRNAs in some tomato tissues suggested their special roles in these tissues' organogenesis and development. Temporal analyses also revealed that the expressions of these miRNAs and mRNAs were highly regulated by different viral infections. Taken together, the observed spatially and temporally changes in miRNAs and target mRNAs expression levels indicate the abilities of different viruses to interfere with miRNA pathway, and are correlated with their respective functions in phenotype determination in different tomato tissues.

The 3' untranslated region of cucumber mosaic virus (CMV) subgroup II RNA3 arose by interspecific recombination between CMV and tomato aspermy virus.
Recombination in single-stranded RNA viruses is one of the principal mechanisms responsible for their evolution. Here we show, using a variety of different methods, that the 3' untranslated region (3'UTR) of subgroup II strains of cucumber mosaic virus [CMV(II)] is related more closely to that of tomato aspermy virus (TAV) than to those of CMV(I) strains. These results suggest that the CMV(II) ...

RNA 1 of the V strain of tomato aspermy virus (TAV) consists of 3410 nucleotides and contains one open reading frame (ORF) of 2982 nucleotides, resembling RNA 1 of cucumber mosaic virus (CMV) strains Q and Fny (68% and 66% identical, respectively) and of brome mosaic virus (BMV) (41% identical). In comparisons between amino acid sequences, three conserved regions (N-terminal, C-terminal and cen...

RNA 2 of the V strain of tomato aspermy virus (TAV) consists of 3074 nucleotides and contains one open reading frame of 2487 nucleotides. Thus, it resembles RNA 2 of cucumber mosaic virus (CMV) strains Q and Fny (62% identical to both), brome mosaic virus (42% identical) and cowpea chlorotic mottle virus (40% identical). In comparisons between amino acid sequences, three different regions of si...
متن کاملEvaluation of potential risks associated with recombination in transgenic plants expressing viral sequences.
Virus-resistant transgenic plants have been created primarily through the expression of viral sequences. It has been hypothesized that recombination between the viral transgene mRNA and the RNA of an infecting virus could generate novel viruses. As mRNA/viral RNA recombination can occur in virus-resistant transgenic plants, the key to testing this risk hypothesis is to compare the populations o...

Changes in the activity of antioxidant enzymes, proline, and total soluble carbohydrates (TSC) were evaluated in tomato (Solanum lycopercicum cv. Superchief) plants in response to salicylic acid (SA) and jasmonic acid (JA) and inoculation with CMV (Cucumber mosaic virus) on days 0, 1, 2, 4, 6, 8, and 15 after virus inoculation (dpi). Results showed that proline content increased in all treatmen...
